The Squadron was constituted as the 65th Troop Carrier Squadron on 7 Dec 1942 and activated and attached to the 403rd Troop Carrier Group on 12 December 1942. The 65th Troop Carrier Squadron was assigned to the Fifth Air Force on 26 July 1943. It was then assigned to the 54th Troop Carrier Wing on 12 August 1943. It was assigned to the 433rd Troop Carrier Group on 9 November 1943. It was assigned to the 403rd Troop Carrier Group on 20 February 1945.
The 4th Glider Section of the 54th Troop Carrier Wing arrived at Lae in New Guinea on 12 November 1944 and Hollandia on 14 November 1944 aboard USS Latimer. The 4th Glider Section went ashore via LCI and proceeded to the 22nd Replacement Centre. They flew to Biak via C-47s on 16 November 1944 where they were reallocated to the 65th Troop Carrier Squadron.

| 27 Feb 1945 | Morotai |
| 15 Jul 1945 | Dulag, Leyte |
Operated C-47s 1943 to 1945 and C-46s 1944 to 1945.
